I noticed that if you aren't careful u can lose a character. Is there a way to lock a saved game? moving the save file into a different folder perhaps? I really dont wana turn auto saves off unless i have too...

Do a full save. It will never be overwritten.

On 360 at least, when you create a manual save file, it is a completely seperate save file from your autosave.. I assume you are talking about the PC version though, and in that case, I really am not certain.. i'd assume it works the same way however...

YUpp, manual saving, but if there's an auto-save (if I'm not mistaken, the game always keeps the latest three of them) you want to keep and cannot manually save the situation any longer, you can copy the file. I usually copy such stuff to another user's folder or to an usb stick, but I'm on PS3, so I'm not sure how it works on X360. On PC, it should be no problem at all to copy the file into a different folder.
