Yes you can do both. But be warned.

Doing any missions for Granny Rags creates an encounter later in the game, that makes Ghost and Shadow very hard (if not impossible) to get.

It's ok man I'm on my 2nd playthrough.

I've got Ghost, Mostly Flesh and Steel, Shadow, Clean Hands etc..
