So what skills should I focus on if I'd like to pull this off? And where can I find good quantities of Moon Sugar and Skooma?

Definitely sneak. Smugglers, especially Khajiit, tend to be thieves.
Go sneak, lockpick,pickpocket,ect.

Search bandit caves for skooma. If you're lucky, you might run into a skooma dealer on the road.

Khajiit Caravans carry plenty of moon sugar. Riften has plenty of Skooma and moon sugar if you know were to look.

Pickpocket or Lockpick.
What smuggler can't pick locks, or steal from people?
AND ALCHEMY!!!!! You can make Skooma i believe.

Skooma is no longer a drug, it's a minor stamina potion.

That made me sad when I saw that a couple days ago. I had 2 skooma in my inventory that I had forgot about and no one noticed and vendors were not opposed to selling to me. I drank them both after I saw the effects. Im hoping a modder will make an adjustment to those pots and make them more illegal.
