also i have an amulet of 100% resist disease should i disenchant it so i learn the effect or is it better to just have it in the case of random vampire attack ?

As for the amulet, the magnitude of the effect doesn't matter in disenchanting, so keep that one and disenchant something that has less effective resist disease instead.

The "room" with the mask wall is right in front of the entrance to the Labryinth in Labryinthian, the room is to the left of the entrance to the dungeon where you encounter Morokei. The room is very similiar to the entrances to many of the burrows, like Shrouded Heath Barrow near Ivarstead, or the upper section of Silent Moons Camp.
