Okay, so I've been playing Skyrim for about 65+ hours. But I don't want to run into a village killing everyone incase I kill a quest giver. If a quest giver dies, does it mean you can't get the quest ever again?

Yes, if you kill someone who gives you a quest, you wont be able to get that quest.
