If you side with the Volkihar Vampire Clan, Harkon is the leader, until the end of the Main Dawnguard Quest line, then you'll become the leader.

If you side with the Dawnguard, Harkon is the leader, but is killed at the end of the quest line, and no one becomes the leader.
