I wanted to do a complete destruction play-through, where after killing all possible NPCs I would blow up the strip.

whilst there's only one people you can't kill i bet the quest givers and some other people (like benny+mr house) are going to ALOT hard to kill that the average wastelander

Anyways, I think you can't kill Doc Mitchell because he has to guide you through character creation.

Yeah I keep getting the nagging feeling it's Doc Mitchell and that he's gonna pull a vanishing act after character creation. Really don't want it to be him (one of my chars stories starts with popping him) but he seems likely.
House is of course another possibility, he may simply be inaccessible.

Other than that I can't think of anyone who could be 'essential'. I'm just glad they got rid of it, hated it in FO3 and Oblivion.

My theory too, ( Even though I stole it from Gabriel) It would make a lot more seance, They've said he's modeled after Howard Hughes, who went insane from Being Alone. I don't think House would be a zax, because in the achievement picture It does show a picture of House, where the last ZAX I saw, didn't. And for people who say he's an android, It's been mentioned that he was a scientist that specialized in robotics, and he froze himself, to be woken up 200 years later.
