I am aware that there are several .ini files that can be tweeked. What I am not sure of is the locations of these .ini files.

Q1. What directories (If you installed default locations) are these located in?

Q2. If you want to turn off vsync, do you need to modify the settings in more than one location of the .ini files?

Q3. If you want to turn off mouse acceleration, do you need to modiff the settings in more than one location of the .ini files?

Q4. If you want the .ini files to be recreated when you initiate the game, is there more than one location of .ini files that you have to delete?

1) There are 2 directories that have information.
The general game folder. Where the game is installed.
Normally this is in your Steam folder. That is somewhere like ......\Valve\Steam\SteamApps\common\skyrim
Usually on your C: disk in the Program Files folder.

This is where the game lives. Where you install Mods and texture packs, etc.
The default settings are taken from there. When you reset your settings to the original setting.
Those settings are defined in low.ini, medium.ini, high.ini and VeryHigh.ini.
Do NOT edit these files. If you do, you can not get the original defaults back.

The other folder is the player specific folder.
This is in your home folder. The home folder is usually C:\Users\.
Your name could sometimes also be Administrator.
The folder with Skryim stuff is in the "My Documents\My Games\Skyrim" folder.
This folder holds 2 ini files. Skyrim.ini and SkyrimPrefs.ini.
Open them with NotePad or another text-editor to look what is where.
Skyrim.ini has only a few settings. SkyrimPrefs.ini has a lot more.
Also, here is the folder Saves, with all your savefiles. In case you want to copy them.

2) If you really want to turn v-sync on or off, it's much better to do it via your videocard driver.
Let us know if you need help to do that. You need at least tell us what brand videocard you have.
Google should show it to you too.
There is an option in the ini files to do something what people often call "turning v-sync off".
That is the option iPresentInterval=1 in SkyrimPrefs.ini.
I run with v-sync enabled in the control panel. And I set iPresentInterval=1 to 1. With it set to 0, the game feels slightly choppier. This is both with a gtx260 (my old card) and a gtx680 (my new card).

3) Can't tell you about mouse acceleration for sure. Sorry.
I have it set to bMouseAcceleration=0. I believe that means off. I think that's the default.
I don't think I have changed it. I don't think there are more places where you need to edit it.
In general, you make changes to ini files once. Either in Skyrim.ini or in SkyrimPrefs.ini.

4) Never delete the files in your system's folder. Only in the user's folder. (See answer #1).
I believe if you delete your Skyrim.ini and SkyrimPrefs.ini files, the next time you start the game, the settings
from one of the low/medium/high/veryhigh.ini files is copied to create a new file.

Thanks for all the info. I think my question #2 and #3 were a little vague. What I have found are the two .ini files mentioned above in my games folder. And I only have one in the system folder, which is the SkyrimPrefs.ini. I was unsure if I had to modify the SkyrimPrefs.ini in BOTH locations for the vsync and mouse acceleration tweeks, or just in one location? If I am to modify the SkyrimPrefs.ini file, I'd just like to confirm that I'd be modyfying the one in the games folder and not the one in the system folder?

Looks like you have vsync turned on via control panel and also in your ini file. I will look into turning off vsync via the video settings.
